2003年加拿大圭尔夫大学的Paul Hebert等人提出了DNA条形码的概念,他们通过对动物界中11门13320个物种分析后,正式提出将一段长648bp的线粒体细胞色素C氧化酶亚基I(cytochromecoxidaseI,COI)基因作为动物鉴定的通用条形码序列。由于传统分类学不能满足现代物种鉴定的需求,DNA条形码技术利用较短的基因保守序列进行种的鉴定,可以在更大范围应用于生态保护研究。DNA条形码自提出以来,受到了世界各国分类和鉴定学家的高度重视,Nature发表了题为“DNA barcoding, a useful tool for taxonomists”的文章,文中指出DNA条形码是国际上近年来发展起来的物种鉴定新技术。Miller在PNAS上发表的文章中提出DNA条形码技术正在推动分类学的“文艺复兴”。DNA条形码技术摆脱了传统形态鉴定方法依赖长期经验的障碍,通过建立标准数据库,可实现快速准确的鉴定,是分子鉴定方法学上的创新。
